Design: Frozen stocks of Legionella were cultured on buffered-charcoal yeast-extract (BCYE) agar and incubated at 37C to obtain isolated colonies for DNA extraction. Nucleic acids from both clinical and environmental samples of Legionella were extracted for WGS with a modified Masterpure DNA Isolation Kit procedure (Epicentre, Madison, WI, USA) . Extracted DNAs were diluted to a final concentration of 2ng/_L before WGS submission.

We investigated the value of whole-genome sequencing (WGS) and single nucleotide polymorphism (SNP) analyses in determining the relationships among and evolutionary rates of Legionella species with long-term persistence in three healthcare facilities. We examined retrospective clinical and environmental isolates of Legionella micdadei and Legionella pneumophila serogroup 1 isolates with identical PFGE DNA fingerprints sampled over the course of up to 18 years.
